Extracellular matrix: Composition and Remodelling

ثبت نشده
چکیده

Introduction The extracellular matrix is a network of proteins and carbohydrates that supports and surrounds the cells in connective tissues. In addition to their structural-mechanical functions ECM molecules regulate diverse cellular functions, and play a significant role in several physiological and pathological processes, such as embryonic development, inflammation and wound healing, hemostasis, fibrosis of various organs, tumor growth and metastasis, and angiogenesis. The interaction of ECM molecules with the cells is conducted through cell-matrix receptors including integrins (see. the seminar material on Cell adhesion). The extracellular matrix is not static: it is remodelled constantly, which implies its constant breakdown by proteases, notably matrix metalloproteinases. During the seminar, we will discuss this topic according to the following scheme:

Changes in the extracellular matrix during myocardial remodelling

The present article focuses on the pathophysiology of myocardial extracellular matrix (ECM) remodelling in chronic heart failure (CHF). The trigger mechanisms for ECM remodelling include acute loss of contractile elements as a consequence of myocardial ischaemia, mechanical tension, oxidative stress and neurohumoral activation. The ECM phenotype changes as a result of the expression of fetal re...

Myocardial extra-cellular matrix and its regulation by metalloproteinases and their inhibitors.

Cardiovascular disease poses a major health care burden in the Western world. Following myocardial injuries, ventricular remodelling and dysfunction ensue, which can eventually culminate in heart failure. An important event in left ventricular (LV) remodelling is alteration of the extracellular matrix (ECM) integrity, the structural network that interconnects the myocardial components. The crit...

Quantitative analysis of 3D extracellular matrix remodelling by pancreatic stellate cells

Extracellular matrix (ECM) remodelling is integral to numerous physiological and pathological processes in biology, such as embryogenesis, wound healing, fibrosis and cancer. Until recently, most cellular studies have been conducted on 2D environments where mechanical cues significantly differ from physiologically relevant 3D environments, impacting cellular behaviour and masking the interpreta...

Extracellular matrix remodelling in response to venous hypertension: proteomics of human varicose veins

AIMS Extracellular matrix remodelling has been implicated in a number of vascular conditions, including venous hypertension and varicose veins. However, to date, no systematic analysis of matrix remodelling in human veins has been performed. METHODS AND RESULTS To understand the consequences of venous hypertension, normal and varicose veins were evaluated using proteomics approaches targeting...

Matrix metalloproteinases and tissue inhibitors of metalloproteinases in ovarian function.

Growth and ovulation of follicles, and development, maintenance, and regression of the corpus luteum depend on cyclical remodelling of the extracellular matrix. The extracellular matrix consists of proteinaceous and non-proteinaceous components and provides the tissue-specific, extracellular architecture to which cells attach, and modulates the activities of cells through cell surface receptors...
